Cílem této práce bylo stanovit vhodné podmínky pro indukci bioluminiscence této bakterie, které by umožnily vývoj bioassay nebo konstrukci biosenzoru k detekci rtuti. MSM (Mineral salt medium) se ukázalo pro indukci bioluminiscence vhodnější než PBS (Phosphate buffered saline). Dosažená maxima bioluminiscence byla vyšší a závislost na koncentraci byla použitélná pro kalibraci. Naopak v PBS způsobovaly vyšší koncentrace rtuti inhibici bioluminiscence. Ukázalo se, že třepání výrazně ovlivňuje bioluminiscenci, vzorky je vhodnější třepat. Měření se suspenzí buněk a půdy vykazovalo hodnoty bioluminiscence blízké pozadí, což se bylo pravděpodobně způsobeno silným zákalem a absorpcí světla emitovaného buňkami. Proceduru bude proto nutné obohatit o extrakční krok. Výsledky ukazují, že je možné použití bioreportétu E.coli ARL1 pro biologickou detekci a stanovení rtuti v půdě, nicméně další výzkum, který by vedl k ucelené podobě bioanalytické procedury, je nutný. Escherichia coli ARL1 is a genetically engineered microorganism, harboring the mer-lux genetic construct . The bioreporter produces bioluminescence in the presence of mercury ions. The aim of this study was to determine the appropriate conditions for the induction of bioluminescence of this strain, which would allow the development of a bioassay or construction of a sensitive biosensor for detection of mercury. MSM (Mineral Salt Medium) medium was found to be more appropriate than PBS (Phosphate Buffered Saline). Detected maxima of bioluminescence were higher and the concentration dependence was applicable for calibration. In contrast, in PBS higher concentrations of mercury caused inhibition of bioluminescence. Shaking greatly influenced the bioluminescence, therefore it is more appropriate to shake the samples. Measurement of a suspension of cells and soil exhibited low bioluminescence close to background. This was probably caused by strong turbidity and absorption of light emitted by the cells. The procedure will have to be enhanced by the extraction step. The results show that application of the bioreporter E. coli ARL1 for biological detection and determination of mercury in the soil is possible, however it further research is needed to achieve a comprehensive form of bioanalytical procedures.
